Zusammenfassung: | Provided is a genetically-modified human pluripotent cell line in which a Brachyury gene and a fluorescent reporter gene are co-expressed, by inserting an insertion sequence containing the nucleotide sequence of the fluorescent reporter gene after a stop codon of the Brachyury gene. A differentiation process when differentiating a cell line into the mesoderm can be monitored by observing fluorescent signals, through performing genetic modification so that the fluorescent reporter gene is expressed by the expression of the brachyury gene, which is a mesodermal marker.
브라큐리 유전자의 정지코돈 뒤에 이어서, 형광리포터 유전자 염기서열을 포함하는 삽입서열이 삽입되어 브라큐리 유전자 및 형광리포터 유전자가 함께 발현되는 유전자 조작된 인간 전분화능 세포주를 제공한다. 중배엽 마커인 브라큐리 유전자의 발현에 의해 형광리포터 유전자가 발현되도록 유전자 조작을 하여 세포주를 중배엽으로 분화시킬 때 형광 시그널을 관찰함으로써 분화 과정을 모니터링할 수 있다. |
