Zusammenfassung:The present invention relates to an RNA expression regulating or editing method through chemical or optogenetic activity control of Cas13 protein. Specifically, in order to control the activity of Cas13 in a CRISPR-Cas13 system, Cas13 protein fragments are generated so as to be recombinable, and a chemogenetically or optogenetically linkable protein is linked to each fragment so that treatment with a low-molecular-weight compound or light irradiation was identified to activate the Cas13 protein. Accordingly, the method of the present invention can regulate the expression of a disease-related RNA or edit RNA mutants, thereby being advantageously used for treating diseases. 본 발명은 화학적 또는 광유전학적 Cas13 단백질의 활성 조절을 통한 RNA 발현 조절 또는 편집 방법에 관한 것으로, 구체적으로 CRISPR-Cas13 시스템에서 Cas13의 활성을 조절하기 위하여, 재결합이 가능하도록 Cas13 단백질의 절편을 생성하고 화학유전학적 또는 광유전학적으로 결합 가능한 단백질을 각 절편에 연결하여 저분자 화합물의 처리 또는 광 조사로 Cas13 단백질이 활성화될 수 있음을 확인하였다. 이에 본 발명의 방법은 질병과 관련된 RNA 발현 조절 또는 RNA 돌연변이를 편집하여 질병 치료에 유용하게 사용될 수 있다.
