Zusammenfassung:The present invention relates to a DNA polymerase variant that belongs to family A and a use thereof. More specifically, the present invention relates to a DNA polymerase variant which easily undergoes polymerization when the base at the 3' end of a primer is complementary to a template, and yet inhibits polymerization when the base at the 3' end of the primer is non-complementary to the template, thereby facilitating the discrimination between the two cases, a PCR process using the same, and a PCR kit comprising the same. The present invention is useful for genotyping of single nucleotide polymorphism (SNP), somatic mutation detection, and the like. 본 발명은 패밀리 A에 속하는 DNA 중합효소 변이체 및 그 이용에 관한 것으로서, 프라이머 3' 말단의 염기가 주형과 상보적일 때는 중합이 원활하게 일어나며, 비상보적일 때는 중합이 억제되어 두 경우의 구별 (discrimination)이 용이하게 되는 DNA 중합효소 변이체, 이 변이체를 이용한 PCR 방법 및 이 변이체를 포함하는 PCR 킷트에 관한 것이다. 본 발명은 단일염기다형성 분석 (SNP genotyping) 및 체세포 돌연변이 검출 (somatic mutation detection) 등에 유용하다.
