Zusammenfassung:The present invention relates to a method for converting a buffer through two-stage diafiltration and, more specifically, to a method for diafiltering a solution containing a protein, a method for preparing a protein preparation by using the same, and a protein preparation obtained using the same. The present invention enables fast and effective progress for the establishment of buffer conversion and concentration processes of a sample containing a protein and enables the effective production of a protein drug formulation with a desired buffer in a desired concentration. Additionally, the present invention has small variations in the pH for each stage, thereby being effective in maintaining the stability of a protein. 본 발명은 2 단계의 투석 여과를 통한 버퍼 변환 방법에 관한 것으로서, 더욱 상세하게는 단백질을 포함하는 용액의 투석 여과 방법, 이를 이용한 단백질 제제의 제조 방법 및 이를 이용한 단백질 제제에 관한 것이다. 본 발명은 단백질을 함유하고 있는 시료의 버퍼 변환 및 농축 공정을 설립하는데 빠르고 효과적으로 진행 할 수 있으며, 단백질 의약품 제형을 원하는 버퍼와 원하는 농도로 효율적으로 생산할 수 있다. 또한, 각 단계별로 pH 변동성이 적어 단백질 안정성을 유지하는데 효과적이다.
