Zusammenfassung: | The present invention relates to a composition for enhancing migration or differentiation of stem cells, comprising colony stimulating factor-2 (CSF-2) protein as an effective component, and to a method for culturing stem cells with enhanced migration and differentiation potentials. In the present invention, it was confirmed that the CSF-2 protein, when used to treat adult stem cells, adipose-derived mesenchymal stem cells, increases stemness, and that when culturing by adding the protein to an osteoclast differentiation culture, osteoblast differentiation potential is greatly enhanced. In addition, it was confirmed that the expression of EMT markers including fibronectin (FN 1), snail, twist, and vimentin is significantly increased, and thus greatly improves migration potential, and at the same time, that cytoskeletal proteins of stem cells are rearranged into a spindle shape. Thus, the CSF-2 protein can be beneficially used as a composition for enhancing the therapeutic effect of stem cell medicine.
본 발명은 CSF-2(colony stimulating factor-2) 단백질을 유효성분으로 함유하는 줄기세포의 이동 또는 분화 개선용 조성물, 이를 이용한 이동 및 분화 능력이 개선된 줄기세포 배양 방법에 관한 것으로, 성체 줄기세포인 지방유래 중간엽 줄기세포주에 CSF-2 단백질을 처리하였을 때 줄기세포성(stemness)을 증가시키고, 조골세포 분화 배지에 상기 단백질을 첨가하여 배양하였을 때 조골세포(osteoblast)로 분화(differentiation)하는 능력이 크게 향상된 것을 확인하였으며, EMT 마커(marker) 유전자들인 피브로넥틴 1(fibronectin, FN 1), 스네일(snail), 트위스트(twist) 및 비멘틴(vimentin)의 발현이 현저히 증가하여 세포 이동(migration) 능력이 크게 향상됨과 동시에, 줄기세포의 세포골격 단백질(cytoskeletal protein)이 방추형(spindle shape)으로 재배열(rearrangement)되는 것을 확인함으로써, 상기 CSF-2 단백질은 줄기세포 치료제의 치료효과를 향상시키기 위한 조성물로 유용하게 사용될 수 있다. |
