Zusammenfassung: | The present invention relates to a method for producing a gene treated induced pluripotent stem cell from a patient-derived somatic cell strain through a one-step method combined with an induced pluripotent stem cell (iPSC) production factor and a gene treatment factor. A method for producing an iPSC having genetic deformation through a one-step scheme may produce a gene-corrected iPSC or a mutation-induced disease model iPSC by simultaneously introducing a reprogramming episome vector for inducing counter differentiation of an adult somatic cell and a gene-corrected or mutation-induced carrier to a host cell. Accordingly, the gene-corrected or mutation-induced iPSC exhibits the same and different properties as and from different kinds of normal-type iPSCs, so the gene-corrected or mutation-induced iPSC may be used in screening a disease treating cell treating agent or a treating material.
본 발명은 유도만능줄기세포(induced pluripotent stem cell, iPSC) 제작 요소 및 유전자 치료 요소를 결합한 원-스텝(one-step) 방법을 통해 환자 유래 체세포주로부터 유전자 치료된 유도만능줄기세포를 제작하는 방법에 관한 것으로, 상기 원-스텝(one-step)에 의해 유전적 변형을 갖는 유도만능줄기세포(induced puripotenet stem cell, iPSC) 제조방법은 성체 체세포의 역분화를 유도하는 리프로그래밍 에피좀 벡터, 및 유전자 교정 또는 돌연변이 유발 전달체를 동시에 숙주세포로 도입하여 유전자 교정된 iPSC 또는 돌연변이 유발된 질환모델 iPSC를 제조할 수 있고, 상기 유전자가 교정되거나, 돌연변이가 유발된 iPSC가 다른 종류의 정상형의 iPSC와 동일한 특징 및 상이한 특징을 나타냄으로써 질환치료용 세포치료제 또는 치료물질의 스크리닝에 사용할 수 있다. |
