Zusammenfassung: | The present invention relates to reverse transcriptase with improved thermostability and more specifically, to mutant reverse transcriptase with improved thermostability which is obtained by substituting more than one amino acid in the amino acid sequence of M-MLV derived transcriptase of sequence number 1 which is selected from glutamine (Q63) in 63^th of the amino acid sequence, lysine (K264) in 264^th of the amino acid sequence, lysine (K295) in 295^th of the amino acid sequence, threonine (T306) in 306^th of the amino acid sequence, glutamic acid (E346) in 346^th of the amino acid sequence, proline (P408) in 408^th of the amino acid sequence, histidine (H438) in 438^th of the amino acid sequence, and asparagine (N454) in 454^th of the amino acid sequence with another amino acid. The mutant reverse transcriptase has excellent thermostability compared to wild-type reverse transcriptase, and offers stable reverse transcription activity for users to obtain cDNA when the users cannot effectively obtain reverse transcription results due to the structural interference of specific RNA capable of forming a stable secondary structure at a high temperature. |
