Zusammenfassung:PURPOSE: A method for preparing GABA(gamma-aminobutyric acid) from novel glutamate decarboxylase(GAD) and high concentration L-glutamate is provided to obtain salt-free gamma-aminobutyric acid. CONSTITUTION: A glutamate decarboxylase mutant is prepared by deleting threonine or adding alanine at carboxy terminal in an amino acid sequence of sequence number 1. The mutant has an affinity tag at an amino terminal. The affinity tag is histidine, maltose binding protein(MBP), thioredoxin(Trx), NusA(N utilization substance A), or glutathione-S-transferase(GST) tag. A method for preparing GABA comprises a step of contacting glutamate or a glutamate salt with the glutamate decarboxylase mutant. The glutamate decarboxylase mutant is fixed on a solid support.
