Zusammenfassung: | PURPOSE: A nucleotide sequence and an amino acid sequence of deoxy-scyllo-inosose synthase are provided. The enzyme is used for biosynthesis of gentamicin, and mass-produces gentamicin from a microorganism containing the deoxy-scyllo-inosose synthase gene. CONSTITUTION: A nucleotide sequence of deoxy-scyllo-inosose synthase is set forth in SEQ ID NO: 1. An amino acid sequence of deoxy-scyllo-inosose synthase is set forth in SEQ ID NO: 2. A microorganism producing deoxy-scyllo-inosose synthase is produced by transforming a microorganism with an expression vector containing the nucleotide sequence of deoxy-scyllo-inosose synthase of SEQ ID NO: 1. A method for producing recombinant deoxy-scyllo-inosose synthase comprises culturing the transformed microorganism and recovering the recombinant deoxy-scyllo-inosose synthase from the cultured medium.
본 발명은 젠타마이신 생합성에 관여하는 효소의 유전자 염기서열 및 아미노산 서열에 관한 것으로서, 좀더 자세히는 마이크로모노스포라 퍼퓨리아(ATCC 15835)로부터 유래한 디옥시실로이노소스 합성효소(deoxy-scyllo-inosose synthase)의 유전자 염기서열과 아미노산 서열을 결정하여 제공한다. 상기 디옥시실로이노소스 합성효소는 아미노글라이코사이드계 항생제의 핵심 구성요소인 2-deoxystreptamine(2-DOS)를 생합성하는 경로에 매우 중요한 효소이므로, 상기 효소를 암호화하고 있는 유전자를 아미노글라이코사이드계 항생제 생산균주에 도입하여 얻은 형질전환체로부터 다량의 항생물질을 생산할 수 있다. |
