ANTI-ROTAVIRUS and IgY/EGG CONTAINING ANTI-ROTAVIRUS and METHOD OF PREPARING IgY/EGG CONTAINING ANTI-ROTAVIRUS

PURPOSE: An anti-rotavirus antibody, immunoglobulin yolk/egg(IgY/egg) containing the anti-rotavirus antibody, and a method for preparing the IgY/egg are provided, thereby producing IgY/egg having high titer and effectively inhibiting rotavirus. CONSTITUTION: An anti-rotavirus antibody composition shows specificity to a rotavirus antigen VP8, wherein the anti-rotavirus antibody composition further shows specificity to a rotavirus antigen VP5 and one or more antigens selected from rotavirus antigen VP4, rotavirus antigen VP7 and rotavirus; and the rotavirus is Rotavirus KRV01(KGCC-11313). A method for preparing IgY/egg containing the anti-rotavirus antibody comprises the steps of: culturing rotavirus; preparing cDNA corresponding to an antigen VP8 from rotavirus RNA; preparing dsDNA from the cDNA; digesting dsDNA and inserting the digested dsDNA into a plasmid; inserting the plasmid into E. coli; expressing the antigen gene in the plasmid to produce an antigen; administrating the antigen into a laying chicken; and obtaining IgY/egg from the immunized chicken. 본 발명은 로타바이러스에 대한 재조합 항원을 생산하고 이를 이용하여 항 로타바이러스 항체 조성물과 이를 함유하는 면역난황/면역란 및 이들의 생산방법에 관한 것이다. 보다 구체적으로는 한국인 유아로부터 분리한 로타바이러스로부터 VP8과 VP5 유전자를 얻어 이를 대장균에 재조합함으로써 VP8과 VP5 단백질을 대량으로 생산하고, 이를 항원으로 사용하여 산란계로부터 항 로타바이러스 항체 및 로타바이러스 항체가 함유된 면역난황/면역란을 생산하여, 유아의 설사 원인인 로타바이러스에 대한 예방의학적 기능성 소재를 생산하는 방법에 관한 것이다. 이러한 본 발명은, 한국형 로타바이러스 재조합 항원을 생산, 2가지 이상의 항원으로 조합 백신을 제조하여 광범위, 고역가의 항 로타바이러스 난황 항체를 생산하였고, 이를 이용하여 로타바이러스를 억제 할 수 있는 식품을 제조할 수 있는 효과가 있다.
