Zusammenfassung: | PURPOSE: A carboxyltransferase of human acetyl-coA carboxylase 2 and a preparation process thereof are provided, thereby improving the preparation yield and purity of carboxyltransferase. CONSTITUTION: A carboxyltransferase of human acetyl-coA carboxylase 2 has the amino acid sequence of SEQ ID NO: 2, wherein the carboxyltransferase comprises amino acids positioned at 1972 to 2359 in the amino acid sequence of human acetyl-coA carboxylase. A polynucleotide encoding the human carboxyltransferase has the nucleotide sequence of SEQ ID NO: 1. An expression vector containing the polynucleotide of SEQ ID NO: 1, wherein the expression vector is pET21b-CT. A transformed microorganism is produced by transformation with the expression vector pET21b-CT, wherein the transformed microorganism is E. coli BL21(DE3). A method for preparing the carboxyltransferase of human acetyl-coA carboxylase 2 comprises the steps of: culturing E. coli BL21(DE3); destroying the cultured E. coli BL21(DE3) in a buffer solution; and subjecting the centrifuged supernatant to the Ni-NTA(nickel-nitrilotriacetic acid) column, anion exchange resin column, and gel-filtration column.
