Zusammenfassung: | PURPOSE: A bicistronic expression vector for animal cells, a transformed cell line containing the same vector and a method for producing antibody proteins using the same are provided, thereby effectively expressing various genes such as an antigen gene. CONSTITUTION: A bicistronic expression vector for animal cells comprises a cytomegalovirus(CMV) promoter required for producing the transcriptional RNA, at least one foreign gene selected from light chain gene or heavy chain gene of an antibody, an IRES sequence, a selective gene and a poly(A) sequence, sequentially, wherein the selective gene is dihydroxyfolate reductase(DHFR) or neomycin resistance gene(NEOR); the bicistronic expression vector is selected from pLCM-MoK(KCTC 10151BP), pLCD-MoK(KCTC 10151BP), pLCN-MoH(KCTC 10150BP) and pANTIBODY(KCTC 10152BP). A transformed animal cell is produced by transformation with the bicistronic expression vector. A method for producing the foreign gene selected from the light chain or heavy chain of the antibody is characterized by culturing the transformed animal cell.
본 발명은 동물세포에서 바이시스트로닉 발현벡터(bicistronic expression vector)를 이용하여 선택용 표지유전자로 선택시 또는 증폭용 유전자를 증폭할 때 목표유전자를 효율적으로 증폭시킴으로써 원하는 단백질을 고농도로 발현시키는 동물세포용 발현벡터, 이를 포함하는 동물세포주 및 이를 이용한 단백질의 생산방법에 관한 것으로, 항체유전자를 비롯한 여러가지 유전자를 효율적으로 발현시키는데 사용할 수 있다. |
