Zusammenfassung: | PURPOSE: An in-vitro culturing method for fertilized eggs and use thereof are provided, which enhances survival rate of fertilized egg from 7.2% to 16.7%(more than 130%), and enhances normal calf delivery rate from 56.2% to 82.6%(more than 50%), and which enables fertilized eggs to produce transformed milk cow when transplanting by melting after freeze-drying preservation, so you can preserve fertilized eggs in-vitro for a long time. CONSTITUTION: The in-vitro culturing method for fertilized eggs comprises: in-vitro maturing a non-fertilized eggs from female cow in a medium; preparing sperms by melting and diluting iced cow sperms after centrifuging; in-vitro fertilizing by mixing and culturing the non-fertilized eggs and the sperms; preparing representing vector by treating pBL-1 with restriction enzyme SacI and SaII, and microinjecting the vector into in-vitro fertilized eggs; preparing mouse embryo fibroblast by Joyner's method, and pouring the fertilized eggs in a CR1aa medium containing the monolayer of embryo fibroblast, and culturing for 5days at 38.5°C, 5% CO2, culturing again in another medium, and examining the growth rate using a microscope. |
