Zusammenfassung: | PURPOSE: A process for reproducing a human granule-colony stimulating factor(hG-CSF) is provided which can reduce a reaction volume and obtain a high yield of the hG-CSF by using an inclusion body of the hG-CSF, so it is easy to produce the hG-CSF in a great quantity CONSTITUTION: The process comprises the steps of: suspending the inclusion body of the hG-CSF expressed from a recombinant strain which is cultivated in a solution containing DL-Dithiotheritol(DTT) and sodium dodecyl sulfate(SDS); adjusting pH of the culture solution to an alkali condition; passing the culture solution through a gel filtration column chromatography equilibrated with a buffer solution containing SDS and Cu¬2+; and recovering a fraction of the reproducing hG-CSF.
본 발명은 재조합 숙주세포에서 발현된 봉입체 형태의 인체 과립성 백혈구 콜로니 자극인자를 DL-디티오쓰레이톨(DL-Dithiothreitol, DTT) 및 소듐 도데실 설페이트(sodium dodecyl sulfate)를 함유하는 용액에 현탁시킨 후 pH를 알카리로 조절하여 용해시키는 단계, 소듐 도데실 설페이트(sodium dodecyl sulfate) 및 구리이온(Cu)을 함유한 완충용액으로 평형화시킨 겔 여과 컬럼크로마토그래피를 통과시키는 단계, 재생된 인체 과립성 백혈구 콜로니 자극인자 분획을 회수하는 단계를 순차적으로 수행하는 것을 특징으로 하는 봉입체 형태의 인체 과립성 백혈구 콜로니 자극인자의 재생(Refolding)방법에 관한 것이다. |
