Zusammenfassung: | The present invention relates to a multistage decellularized mammal-derived tissue matrix and a method for producing the same, in which the decellularized mammal-derived tissue matrix is prepared through: a preparation step of preparing tissues of a mammal except for humans; a pretreatment step of pretreating the tissues; a primary inactivation step of using alcohol so as to inactivate viruses included in the pretreated tissues; a primary decellularization step of using an aqueous base solution so as to remove cells from the tissue in which viruses are inactivated; a secondary decellularization step of performing enzyme treatment on the primarily decellularized tissues so as to remove cells; and a secondary inactivation step of using acid so as to inactivate viruses included in the decellularized tissues, wherein an amount of DNA in the tissue having undergone the secondary inactivation step is 50 ng/mg or less and an elastin reduction rate (L) in the tissue having undergone the pretreatment step or the secondary inactivation step is 20% or less. According to the present invention, the tissue matrix has a very low amount of DNA so that immune responses or foreign body reactions are minimized, and has a high amount of elastin so that wound-healing and scar-formation suppression effects are high.
본 발명은, 탈세포화된 포유류 유래 조직 매트릭스에 있어서, 사람을 제외한 포유류의 조직을 준비하는 준비 단계; 상기 조직을 전처리하는 전처리 단계; 알코올을 사용하여 전처리된 조직에 포함된 바이러스를 불활성화하는 제1 불활성화 단계; 바이러스 불활성화된 조직에서 염기 수용액을 사용하여 세포를 제거하는 1차 탈세포화 단계; 1차 탈세포화된 조직을 효소처리하여 세포를 제거하는 2차 탈세포화 단계; 및 산(acid)을 사용하여 탈세포화된 조직에 포함된 바이러스를 불활성화하는 제2 불활성화 단계;를 거쳐 제조되며, 상기 제2 불활성화 단계를 거친 조직의 DNA 함량은 50ng/mg 이하이고, 상기 전처리 단계 내지 제2 불활성화 단계를 거친 조직의 엘라스틴 함량의 감소율(L)이 20% 이하인 것을 특징으로 하는, 다단계 탈세포화된 포유류 유래 조직 매트릭스 및 이의 제조 방법에 관한 것으로, 본 발명의 조직 매트릭스는 DNA 함량이 매우 낮아 면역이나 이물 반응을 최소화하고, 엘라스틴 함량이 높아 상처 치유, 흉터 억제 효과가 높은 장점이 있다. |
