The present invention relates to an agarwood pill composition and a preparation method thereof which comprises: a first step of mixing agarwood powder with a red ginseng concentrate and allulose to prepare and age a primary mixture; a second step of mixing the primary mixture with mixed plant powder to prepare and age a secondary mixture; a third step of adding a microorganism fermented solution and a garlic concentrate to the secondary mixture to prepare and age a tertiary mixture; a fourth step of mixing the tertiary mixture with Maca powder, red ginseng powder, and deer antler powder to prepare and age a final mixture; and a fifth step of kneading the final mixture prepared in the fourth step to prepare a dough and tearing a predetermined amount off from the dough and forming a spherical shape to age the same.
