Zusammenfassung:Provided is a process for preparing N-(trans-4-isopropylcyclohexylcarbonyl)- D-phenylalanine having excellent hypoglycemic action upon oral administration, and useful as an agent for treating diabetes. The process for preparing N-(trans-4-isopropylcyclohexylcarbonyl)- D-phenylalanine represented by the following formula 1 comprises the steps of: reacting trans-4-isopropylcyclohexane carboxylic acid or a compound represented by the following formula 8 with a dialkyl chlorophosphate or O,O-dialkyl chlorothiophosphate (wherein alkyl includes -OCH3, -OC2H5 or -OC6H5) to obtain a compound represented by the following formula 10; reacting the compound represented by formula 10 with a compound represented by the following formula 11 to obtain a compound represented by the following formula 12; and hydrolyzing the compound represented by formula 12 with an acid. In the above formulae, R' is O or S.
