Zusammenfassung: | PURPOSE:To efficiently produce human lysozyme, by transforming an yeast with a recombinant plasmid holding a human lysozyme gene linked to a secretory leader sequence and processing signal sequence. CONSTITUTION:A synthetic oligonucleotide, expressed by the formula linked to alpha-factor gene at the HindIII terminal and to human lysozyme gene at the TaqI terminal is prepared. A DNA fragment containing a synthetic human lysozyme and leader processing signal of an alpha-factor is linked to the above- mentioned nucleotide by treatment with T4 ligase to give a recombinant plasmid pLY26. A DNA fragment containing a selection marker TRP1 and a replication starting point ARS1 is linked to the above-mentioned plasmid to provide a plasmid pLY27, which is then linked to a DNA fragment containing a 2mum plasmid replication starting point to construct a recombinant plasmid pLY28. An yeast NA87-11A strain can be transformed by the above-mentioned plasmid to produce the aimed human lysozyme. |
