Zusammenfassung: | PURPOSE:To use a general-purpose measuring device easy to operate without requiring a long time and to enhance detection sensitivity, by adjusting the concn. of p-hydroxybenzoic hydrazide after the mixing of a reaction reagent solution to 0.05-1wt%. CONSTITUTION:Water or an aqueous salt solution or a liquid mixture thereof with an org. solvent is used as an eluent to separate sugars in a separation column and said saccharides are mixed and reacted with a reaction reagent solution containing p-hydroxybenzoic hydrazide to form a light absorbing substance which is, in turn, detected by a light absorption detector. Herein, the concn. of p-hydroxybenzoic hydrazide after the mixing of the reaction reagent solution is adjusted to 0.05-1wt%. By this method, almost all of the saccharides can be detected stably and easily with high sensitivity. |
