Zusammenfassung: | PURPOSE:To obtain a readily producible monoclonal antibody, reactive with a specific antigen without sialic acid present in an antigenic determinant, having properties of reacting with human cancers in colon, stomach, bile duct, etc., useful for blood serum diagnosis of cancer and belonging to IgM class. CONSTITUTION:A mouse is immunized with an antigen (e.g. colonic cancerous cell) recognizable by a desired monoclonal antibody and antibody-producing cells are separated from the immunized mouse. The separated cells are then fused to myelomatous cells having self propagation ability to afford fused cells capable of producing a monoclonal antibody. The resultant fused cells are subsequently cultivated to provide an antibody, which is recovered to separate a monoclonal antibody, having properties of reacting with a glycoprotein antigen having >=1,000,000mol.wt. (measured by a gel filtration method) in ascitic fluid of a patient suffering from colonic cancer, human colonic cancer, human ovarian mucocele adenocarcinoma, part of human bronchial epithelium, cultivated cell strain of human colonic cancer, human pancreatic cancer and cultivated cell strain of human cholangiocarcinoma H-1, etc., and called NCC-CO-450, etc. |
