Zusammenfassung:PURPOSE:To obtain a recombinant plasmid having high gene amplification capability, by integrating two genetic information sets to regulate biosynthesis of tryptophan into one molecule of a plasmid DNA free from self-replicability in host microorganism. CONSTITUTION:The objective recombinant plasmid contains two fragments of a chromosome DNA of a microorganism (preferably Bacillus amyloliquefaciens) having chloramphenicol-resistant gene and ampicillin-resistant gene and capable of producing L-tryptophan. Each of the fragments contains genetic information essentially participating in biosynthesis of L-tryptophan. The recombinant plasmid is e.g. pSDT391 shown in the figure. E.coli SD1011 containing the pSDT391 has been deposited in the Japanese Fermentation Research Institute as FERM P-8621.
