Zusammenfassung:	PURPOSE:To culture a protoplast separated from a plant of e.g. Brassicaceae family, in high efficiency, division frequency and reproducibility, by embedding the protoplast in agarose, culturing the agarose in a liquid medium free from NH4, and transferring to a liquid medium containing NH4. CONSTITUTION:Protoplast separated from a plant of Brassicaceae family or Solanaceae family is embedded in agarose. The agarose is cultured first in a liquid medium free from NH4, and then in a liquid medium containing NH4 (e.g. NH4NO3, etc.).
