Zusammenfassung: | PURPOSE:To produce a large amount of beta-amylase by a host bacterial strain, by using a passenger DNA originated from Bacillus polymyxa and containing the beta-amylase gene having specific numbers of the recognition site with restriction enzyme HindIII, the recognition site with restriction enzyme HincII, and the recognition site with restriction enzyme Kpn I . CONSTITUTION:A recombinant DNA is prepared by bonding a vector DNA originated from a proper plasmid with a passenger DNA originated from Bacillus polymyxa and containing a beta-amylase gene having 2 recognition sites with restriction enzyme HindIII, 1 recognition site with restriction enzyme HincII and 1 recognition site with restriction enzyme Kpn I . The recombinant DNA is introduced into the cell of a host bacterial strain, e.g. Escherichia coli, Bacillus subtilis, etc., and the strain is cultured in a nutrient medium to produce the objective beta-amylase. |
