Zusammenfassung: | PURPOSE:In the affinity chromatography, a specific serum protein mixture is brought into contact with deactivated thrombin gel to effect simple, rapid and effective removal of fibrinogen with retention of the activity of coagulating VIIIth factor. CONSTITUTION:In the affinity chromatography, gel is brought into contact with a serum protein mixture of 6-8pH and 0.1-1.0 ionic strength to effect specific adsorption and separation of fibrinogen. Namely, a gel containing anhydrothrombin as a ligand is used to pass the protein mixture or stir together with the mixture to separate only fibrinogen with no affect to the VIIIth factor. The process permits the effective separation of fibrinogen from the concentrated preparation for cryoprecipitate and the VIIIth factor to give the drug for treating hemophilia A. |
