CONSTITUTION:Microorganisms belonging to Bordetella pertussis phaseIor II are cultured, and the cultured product, especially the supernatant liquid of the product is treated with a haptoglobin sephalose column, etc. to obtain the objective insulin secretion promoting substance having the following characteristics: molecular weight, 116,000+ or -7,000 (by ultracentrifugation); protein content, >=97wt% (by Lowry method); amino acid composition and ratios (muM/100muM), Asp 7.5-7.9, Thr 6.8-7.8, Ser 5.9-7.6, Glu 8.8-10.0, Pro 5.5-6.4, Gly 8.7-9.6, Ala 9.0-10.8, Cys/21.0-2.1, Val 6.6-7.6, etc.; isoelectric point, 9.1+ or -0.4; specific rotation,[alpha]D =-29+ or -5 deg.; etc. The substance has higher specific activity than the conventional one by a factor of 1.5-3, and is useful as a remedy and preventive of diabetes.
