Zusammenfassung: | PURPOSE:To determine acyl coenzyme A (acyl CoA), etc. and avoid the measurement interference by the clouding of a blood serum, by reacting an acyl CoA synthetase with a sample containing a free fatty acid, CoA, adenosine-5'-triphosphate (ATP) and lipoprotein, and inactivating the CoA, ATP or acyl CoA synthetase, and reacting a lipoproteinic lipase with the resultant sample. CONSTITUTION:An acyl coenzyme A synthetase is reacted with a sample e.g. a blood serum containing a free fatty acid, coenzyme A (CoA), adenosine-5'-triphosphate (ATP) and lipoprotein, to produce an acyl coenzyme A, adenosine-5'- monophosphate and pyrophosphoric acid. The CoA, ATP or acyl coenzyme A synthetase remaining in the reaction solution is then inactivated with an SH reagent, an enzyme capable of decomposing the ATP, a chelating agent, and a lipoproteinic lipase is then reacted with the reaction solution. The measurement is carried out by determining the acyl CoA or adenosine-5'-monophosphate corresponding to the amount of the free acid in the sample stoichiometrically. |
