Zusammenfassung:	PROBLEM TO BE SOLVED: To perform the exponent amplification of a target nucleic acid sequence using a single primer alone without using two primers as conventional. SOLUTION: This method for preparing a primer for single-unpaired primer exponent amplification process comprises the following steps: (a) selecting a target nucleic acid sequence having 3'-end and 5'-end, (b) preparing a 1st estimated primer with about 10-40 base pairs designed to form a hybrid on a 1st primer site situated on the 3'-end of the target nucleic acid sequence or in the vicinity thereof, wherein the estimated primer bears 3'- and 5'-ends, and (c) performing an amplification step in order to confirm that the estimated primer has effective in single-unpaired primer exponent amplification.
