Zusammenfassung: | PROBLEM TO BE SOLVED: To obtain the subject new gene useful e.g. for developing new varieties of yeast and the analysis of genetic information, comprising a DNA of a pantothenic acid-demanding complementary gene having a specific base sequence, and functioning as a selective marker for Refined Sake Yeast Association No.7 and variants thereof. SOLUTION: This gene is a new DNA of the pantothenic acid-demanding conplementary gene expressed by a base sequence of the formula, derived from Saccharomyces cerevisiae complementing Refined Sake Yeast Association No.7 and (or) variants thereof and can function as a selective marker for Refined Sake Yeast Association No.7 and (or) variants thereof and can efficiently select transformants. This DNA of the pantothenic acid-demanding complementary gene is obtained by extracting chromosomal DNAs from Saccharomyces cerevisiae X2180-1A, preparing a gene library using the chromosomal DNA according to a conventional technique, then, recovering a plasmid from the gene library, transducing the plasmid into Refined Sake Yeast Association No.7 followed by culturing in a medium containing no pantothenic acid and recovering the DNA from developed colonies. |
