Zusammenfassung: | PROBLEM TO BE SOLVED: To efficiently concentrate the bacterium cells and use them for nucleic acid analysis by adding a specified density-lowering agent to a viscous biological sample in a sufficient amount to reduce the density of the viscous sample lower than the density of the bacterium cells, centrifuging and pelletizing the bacterium cells. SOLUTION: A viscous biological sample selected from the group consisting of respiratory samples, respiratory sedimentation, oral cavity juice, vaginal secretion, semen, wound infection liquid and abscess liquid is diluted with a water-soluble density-lowering agent of a (substituted) methyl alcohol or (substituted) propyl alcohol in a sufficient amount to reduce the density of the sample lower than that of the bacterium cells having 0.7-0.9 g/ml density and boiling at a temp. higher than 50 deg.C, centrifuging and pelletizing arbitrary cells in the sample thereby concentrating the bacterium cells. In a preferred embodiment, the viscous biological sample is pretreated with a reagent that inhibits the analysis of nucleic acid. |
