Zusammenfassung: | PROBLEM TO BE SOLVED: To obtain the subject callus and corm having the increasing demand in a market and the high appreciation value, in an equal proliferation rate for a long period in a large quantity by culturing the callus formed on a solid medium from a plant tissue of ixia in a liquid medium. SOLUTION: After a plant tissue of a corm or the like of Ixia viridiflora is washed with a detergent, the sterization treatment is performed, and then the tissue divided into 4-8 pieces is prepared under sterile condition. This tissue is incubated at 25 deg.C in a dark place for 4 weeks upon standing on a solid medium containing sugar and gelan gum to which 0.01-0.5mg/l of α-naphthalene acetic acid and 0.1-5mg/l of benzyl-aminopurine are added as plant hormones to form a callus. Then, after an adventive germ tissue or adventive embryo tissue is formed from the callus, the corm of ixia is obtained by forming the corm from an infant plant affording by rearing these tissues. |
