Zusammenfassung: | PROBLEM TO BE SOLVED: To detect a disease of chrysanthemum stunt viroid (CSVd) with a high sensitivity by preparing cDNA by carrying out reverse transcription reaction using a primer comprising a partial sequence complimentary to a base sequence derived from chrysanthemum stunt viroid (CSVd) by gene amplification. SOLUTION: In a method for detecting chrysanthemum stunt viroid (CSVd) by gene amplification, a leaf of CSVd-infected chrysanthemum is frozen and ground by a mortar and whole RNA is extracted by an ordinary method and a polymer RNA is removed therefrom by 2M LiCl treatment and low molecular weight RNA is recovered from the residual RNA by precipitation by ethanol and reverse transcription reaction is carried out using a primer containing a sequence comprising a continued >=15 base or its variant sequence among base sequences complimentary to a base sequence represented by the formula as a sequence capable of detecting CSVd with a highest sensitivity among the low molecular weight RNA to prepare cDNA and infection due to CSVd for diagnosis, etc., of chrysanthemum stunt disease is detected with a high sensitivity using the resultant cDNA as a template. |
