Zusammenfassung:PURPOSE: To obtain the subject protease specifically existing in the skeletal muscle of a mammalian exhibiting a muscular dystrophy syndrome, and useful for the research of therapeutic medicines for the muscular dystrophy syndrome and for the diagnosis of the syndrome. CONSTITUTION: This new protease is obtained from the skeletal muscle, etc., of a hereditary muscular dystrophy mouse by an ordinary protein purification method, has an amino acid sequence region of Ala-Ile-Ser-Val-Tyr-Lys-Ile and an amino acid sequence region of Gly-Gly-Thr(or Ile)-Ser(or His)-Gly-Tyr-Val- Pro-Tyr-Ala-Xaa-Thr-Gly in regions except the amino-terminated region of the formula (Xaa is the residue of a man-identified amino acid), and has physicochemical properties cited below. (1) The protease has a mol.wt. of approximately 27000 (SDS-polyacrylamide gel electrophoresis method); (2) the protease exhibits a strong degradation activity against C-proteins, and hyrolyzes also denaturated proteins; (3) the optimal pH is approximately 9, and the stable pH range is 4.0-10.2; (4) the protease has the optimal temperature of approximately 30-40 deg.C, and acts in the temperature range of approximately 15-50 deg.C.
