Zusammenfassung: | PURPOSE: To obtain a new determination gene DNA having a specific restriction enzyme map and a mol.wt., used for identifying Pasteurella piscicida strain which is a bacterium causing bacterial nodositas, and enabling to simply and rapidly identify the bacterium causing fish infections disease. CONSTITUTION: This gene DNA has a DNA restriction enzyme map of the formula and a mol.wt. of approximately 0.8kbp, and is used for determining Pasteurella piscicida strain which is a bacterium causing bacterial nodositas. The gene DNA is useful as a reagent for identifying the bacterial kind of a fish disease, enabling to simply and rapidly identify a bacterium causative of an infectious disease due to the bacterium of the fish disease generated in a fish culture pond, etc. The DNA is obtained by culturing Pasteurella piscicida KG 8601 strain in a culture medium, collecting the cultured cells, extracting the chromosomal DNA from the collected cells by a conventional method, treating the extract with a restriction enzyme, ligating the obtained fragments to vectors, transducing the vectors into Escherichia coli, screening the Escherichia coli in the presence of a probe by a colony hybridization method, selecting a strain containing the strain-determining gene, and subsequently recovering the DNA. |
