Zusammenfassung:	PURPOSE:To provide a novel cholesterol.esterase useful for clinical diagnoses, organic synthesis, detergent enzymes, etc., and improved in stability against organic solvents, surfactants, oxidizing agents and modifying agents. CONSTITUTION:A stable cholesterol.esterase in which at least one of the No.163 Asn and No.264 Leu of a cholesterol.esterase of the formula has been replaced with another amino acid such as a neutral amino acid and whose No.176 amino acid is His or an amino acid (e.g. Glu) except the His. A method for producing the stable cholesterol.esterase comprises introducing a mutagenic fragment into a gene coding a cholesterol.esterase having the amino acid sequence of the formula and obtained from a microorganism, and subsequently collecting the stable cholesterol.esterase from the culture product of a microorganism holding a recombinant DNA recombined with the gene.
