Zusammenfassung: | PURPOSE:To obtain a large amount of a human serum album in useful for treating massive bleeding, shock, burn, hypoproteinosis, fatal erythroblastosis, etc., having low tinction by culturing a human serum albumin producing host prepared by gene manipulation under a specific temperature condition. CONSTITUTION:A human serum albumin (HSA) manifestation plasmid having eliminated a 5' noncoding range of a HAS gene is prepared from a gene coding for HSA under of a promoter of yeast of Pichia. The plasmid is transduced into Pichia pastoris GTS115 strain (NRRL deposition No. Y-15851), which is transformed to give a transformant UHG42-3 strain as a HSA producing host. Then the transformant is cultured at 21-29C temperature condition in a medium containing bactopeptone, yeast extract, glucose, etc., to produce the objective HSA useful as a therapeutic agent for massive bleeding, shock, burn, hypoproteinosis, fatal erythroblastosis, etc., in an enlarged proliferation degree and in reduced tinction. |
