Zusammenfassung: | PURPOSE:To obtain a new sorbitol oxidase, capable of acting on D-sorbitol, consuming oxygen and producing hydrogen peroxide, good in stability and useful as a reagent, etc., for measuring the sorbitol content in erythrocytes to be an index of diabetic complications. CONSTITUTION:A microorganism, having the ability to produce a sorbitol oxidase and belonging to the genus Xanthomonas [e.g. Xanthomonas maltophilia TE3539 (FERM P-13296)] is inoculated into a culture medium and cultured at 30 deg.C and the resultant culture solution is used as a seed culture solution and subsequently transferred to a jar fermenter and cultured at 30 deg.C. The obtained culture solution is centrifuged to collect microbial cells, which are then subjected to pressing treatment and centrifugation. A supernatant is subsequently collected and purified by the fractionation with ammonium sulfate, anion exchange chromatography and gel filtration, etc., to afford the objective new sorbitol oxidase capable of acting on D-sorbitol, consuming oxygen and producing hydrogen peroxide. Furthermore, the resultant new sorbitol oxidase is made to act on a sample containing the sorbitol, etc., to measure the amount of the produced hydrogen peroxide. Thereby, the amount of the sorbitol is measured. |
