Zusammenfassung:PURPOSE:To obtain a monoclonal antibody selectively recognizing an antibody Derf II originated from Dermatophagoides fabrinae, belonging to IgG or IgM and useful for measuring an allergen for bronchial asthma, etc., for purifying antibodies, etc., by immunizing a mammalian with the antigen Derf II originated from Dermatophagoides fabrinae and subsequently subjecting the product to various treatments. CONSTITUTION:The production of the objective Derf II-resistant monoclonal antibody comprises gathering Dermatophagoides fabrinae by a saturated salt water floating method, extracting the gathered mites with a phosphoric acid-buffered salt water, precipitating the extract with a 60% saturated ammonium sulfate solution, purifying the supernatant by an anion exchange column chromatography, isolating an antigen Derf II originated from the Dermatophagoides fabrinae, injecting the isolated antibody together with an adjuvant into a Balb/c mouse, finally immunizing the mouse, collecting the splenic cells of the immunized mouse, fusing the collected splenic cell to a mouse myeloma cell, selectively culturing the fused cell in a HAT medium, cloning the obtained hybridoma by a limiting dilution analysis method, transplanting the obtained monoclonal hybridoma into the abdominal cavity of a mouse, collecting the abdominal dropsy accumulated in the abdominal cavity after approximately two week, subjecting the collected abdominal dropsy to a centrifugal treatment and subsequently purifying the treated product by a column chromatography.
