HUMAN LIPOPROTEIN LIPASE-LIKE PROTEIN

PURPOSE:To obtain the title compound having a specific molecular weight, being separated product of a human lipoprotein lipase(LPL), free from LPL activity, having high immune homology with LPL and useful as an antigen for preparation of anti-LPL antibody. CONSTITUTION:A human myelocytic leukemia cell strain THP-1 strain (ATCC-TIB202) is cultured in the presence of 12-0-tetradecanoyl phorbol-13- acetate and/or heparin, in a fetal calf serum-containing medium at 37 deg.C for 7 day and then cultured liquid is centrifuged to separate proliferated cell and the supernatant is passed through a hydroxyapatite column, which is cleaned and then an absorbed substance are eluted with 1M sodium chloride/0.1 M phosphoric acid buffer solution (having pH 7) and purified by gel permeation and immobilized heparin column treatment to provide the objective protein having about 20000 molecular weight, being human LPL decomposition product, having amino acid sequence of N-terminal part expressed by formula I or formula II, having no human LPL activity and being stable even when allowed to stand in a solution containing human serum at 37 deg.C for 60min.
