Zusammenfassung: | PURPOSE:To produce the subject substance useful as a tackifier, taste-improving agent, plasma substitute, etc., in the fields of food and pharmaceuticals in high productivity by culturing a microbial strain of genus Klebsiella in a culture liquid at an initial pH adjusted to basic state and collecting the objective substance from the culture product. CONSTITUTION:A microbial strain belonging to genus Klebsiella [e.g. Klebsiella planticola KPS5003 (FERM BP-3300)] is proliferated on a slant medium composed of lactose, trypticase, potassium dihydrogen phosphate, magnesium sulfate, manganese chloride and agar. The proliferated cell is collected with an inoculating loop, inoculated on the above medium devoid of agar and precultured at 30 deg.C for 1 day by shaking culture. The precultivation product is inoculated to a culture liquid consisting of the above culture medium after adjusting the initial pH to 8.0-11.0 and cultured at 30 deg.C for 5 days by shaking culture. The culture liquid is subjected to centrifugal separation to remove the microbial cells, ethanol is added to the supernatant liquid and the precipitate is separated and dried to obtain the objective BS-1 substance having high viscosity. |
