PACKING AGENT FOR ENZYMATIC ACTIVITY DETERMINATION, ITS USAGE AND SYSTEM THEREFOR

PURPOSE:To obtain the title packing agent capable of determining enzymatic activity quickly, simply and in high accuracy by binding of biotin bound to a substrate to avidin bound to a water-insoluble substrate to immobilizing the former substrate to the latter. CONSTITUTION:Biotin is bound to a substrate for enzyme, whereas avidin is bound to a water-insoluble substrate, and the former substrate is immobilized, via both the biotin and avidin, on the latter substrate, thus obtaining the objective packing agent. The present packing agent can be obtained, for example, through the following process: (A) avidin-immobilized beads obtained by adding epoxy group-modified beads prepared by introducing epoxy group into methacrylate polymer beads to an avidin solution to carry out reaction and (B) a biotin-bound substrate obtained by adding a second substrate to an N- hydroxysuccinimide biotin solution are added to a phosphoric acid buffer solution to bring both the components A and B into mutual contact.
