Zusammenfassung:PURPOSE:To impart a bacterial cell with the capability to produce heat-resistant leucine-dehydrogenase, by introducing a plasmid containing the gene of leucine-dehydrogenase derived from the chromosome DNA of a thermophilic microorganism into a bacterial cell which can be grown at normal temperature. CONSTITUTION:The gene of leucine-dehydrogenase derived from chromosome DNA is separated from the cell of bacterial strain belonging to thermophilic Bacillus genus, e.g. Bacillus stearothermophilus. The gene of the leucine-dehydrogenase and a DNA acting as a vector are digested with a restriction enzyme, and treated with a ligase to obtain plasmid pICRI having the gene of heat-resistant leucine-dehydrogenase. The capability to produce heat-resistant leucine-dehydrogenase can be imparted to a cell by introducing the plasmid to the cell of e.g. Escherichia coli.
