Zusammenfassung: | PURPOSE:To proliferate and thicken small bulbs and proliferate bulbous plants without requiring hands by keeping a culture containing the small bulbs obtained from tissues of bulbous plants in the vapor phase using tissue culture, spraying a liquid culture medium thereon, feeding O2 thereto and carrying out culturing in the vapor phase. CONSTITUTION:Tissual pieces, such as bulb, stem, leaf, petal, anther, filament, ovary, bud, scale or root, are cut out of bulbous plants which are plants of the order Liliiflorae, such as the family Liliaceae, plants of the order Orchidales including the family Orchidaceae, etc. The surfaces of the above-mentioned tissual pieces are then subjected to sterilization treatment with ethanol, etc., washed with sterile water and user for tissue culture. A solid or liquid culture medium normally used for culturing plants is employed for the above-mentioned tissue culture and phytohormones, such as auxin, are then added thereto. The resultant culture containing the small bulbs is kept in the vapor phase and a liquid culture medium is sprayed thereon. Air or O2 is fed to carry out culturing in the vapor phase and proliferate and thicken the afore-mentioned small bulbs. The obtained bulbs can be transplanted to soil with excellent preservation quality without carrying out acclimating treatment, such as temperature control. |
