Zusammenfassung: | To provide a method for highly accurately measuring dialysate endotoxin concentration by using a calibration curve made with water, and kit used for the method.SOLUTION: An endotoxin measuring method includes: after mixing a prescribed amount of specimen with dry power including lysate reagent to prepare first reaction solution, incubating the first reaction solution for a prescribed time; then, under shade, mixing the first reaction solution and the dry powder including luciferase bioluminescence reagent to prepare second reaction solution and measuring a light emission amount of the second reaction solution; and measuring endotoxin concentration of the specimen based on the light emission amount, where the specimen is dialysate or water, and the second reaction solution has sodium ion concentration of 250-390 mM, calcium ion concentration of 0.5-10 mM, chloride ion concentration of 250-361.5 mM and tris concentration of 20-30 mM.SELECTED DRAWING: None
【課題】本発明は、透析液のエンドトキシン濃度を、水で作成された検量線を用いてより精度よく測定するための方法、及び当該方法に使用されるキットを提供する。【解決手段】所定量の被験試料を、ライセート試薬を含む乾燥粉末と混合して第1の反応溶液を調製した後、当該第1の反応溶液を所定時間インキュベートした後、遮光下で、前記第1の反応溶液と、ルシフェラーゼ生物発光試薬を含む乾燥粉末とを混合して第2の反応溶液を調製し、当該第2の反応溶液の発光量を測定し、前記発光量に基づいて前記被験試料のエンドトキシン濃度を測定し、前記被験試料が透析液又は水であり、前記第2の反応溶液が、ナトリウムイオン濃度が250~390mM、カルシウムイオン濃度が0.5~10mM、塩化物イオン濃度が250~361.5mM、及びトリス濃度が20~30mMである、エンドトキシンの測定方法。【選択図】なし |
