Zusammenfassung:To provide a method of enhancing efficiency of a downstream chromatography step for purification of antibody.SOLUTION: A method comprises the steps of: (a) passing a composition comprising an antibody of interest and various contaminants through an ion exchange membrane, followed by passing a buffer having a pH of 1 to 5 pH units different from the pI of the antibody and a low ionic strength therethrough; (b) overloading the ion exchange membrane such that at least one of the contaminants remains bound to the membrane while the antibody of interest is primarily in the effluent; and (c) collecting the effluent from the ion exchange membrane comprising the antibody of interest.
