Zusammenfassung: | To provide a new target analysis method utilizing polarization degree, with which it is possible to improve the accuracy of analysis.SOLUTION: A target analysis method of the present invention includes: a detection step for causing a fluorescence unlabeled aptamer, a fluorescence labeled complementary chain and a sample to coexist and detecting a polarization degree deriving from the fluorescence labeled complementary chain that depends on binding and non-binding of the fluorescence unlabeled aptamer and fluorescence labeled complementary chain, the fluorescence unlabeled aptamer being not labeled by a fluorescence substance, the constitutional unit including a modified base and a natural base, the aptamer binding to a target, the fluorescence labeled complementary chain being labeled by a fluorescence substance, the constitutional unit not including a modified base and including a natural base, the complementary chain binding to the aptamer; and an analysis step for analyzing the target in the sample on the basis of the polarization degree.SELECTED DRAWING: Figure 1
【課題】 偏光度を利用したターゲットの分析方法について、分析精度を向上できる新たな方法を提供する。【解決手段】 本発明のターゲット分析方法は、蛍光非標識アプタマーと、蛍光標識相補鎖とを使用し、前記蛍光非標識アプタマーは、蛍光物質で標識化されておらず、構成単位が修飾塩基および天然塩基を含み、ターゲットに結合するアプタマーであり、前記蛍光標識相補鎖は、蛍光物質で標識化され、構成単位が修飾塩基を含まず天然塩基を含み、前記アプタマーに結合する相補鎖であり、前記蛍光非標識アプタマーと、前記蛍光標識相補鎖と、サンプルとを共存させ、前記蛍光非標識アプタマーと前記蛍光標識相補鎖との結合および非結合に依存する、前記蛍光標識相補鎖に由来する偏光度を検出する検出工程と、前記偏光度に基づいて、前記サンプル中のターゲットを分析する分析工程とを含む。【選択図】図1 |
