Zusammenfassung: | To provide a method for evaluating aging in epidermal tissue by actively inducing cells to a state close to skin physiological aging phenomenon.SOLUTION: A method for evaluating aging in epidermal tissue is realized, in which medium containing a secretory factor of aged fibroblasts is used upon culturing epidermal keratinocytes and a human three-dimensional cultured epidermal model having a differentiation potential so as to positively induce epidermis to a physiologically aged state, and TEWL (water evaporation amount) is used as an index for barrier function deterioration in the reproduced aging model and gene expression level of a keratin-related gene is used as an index for keratinization deficiency.SELECTED DRAWING: None
【課題】皮膚生理的老化現象に近い状態に細胞を積極的に誘導することにより、表皮組織における老化の評価方法を提供すること。【解決手段】分化能を有する表皮角化細胞及びヒト3次元培養表皮モデルを培養する際、老化した線維芽細胞の分泌因子を含んだ培地を用いることにより、表皮を生理的老化状態に積極的に誘導させ、この再現老化モデルのバリア機能低下としてTEWL(水分蒸散量)及び角化不全として角化関連遺伝子の遺伝子発現量を指標とすることを特徴とする、表皮組織における老化の評価方法を可能とした。【選択図】なし |
