Zusammenfassung: | To provide a method for simply and accurately determining the presence of two or more kinds of specific oligonucleotide.SOLUTION: A determination method includes: a process of mixing a probe having a sequence complementary with two or more kinds of specific oligonucleotide and a measurement sample to obtain a mixture; a process of applying voltage between first solution containing the mixture and second solution which are partitioned by a lipid double membrane having a nanopore, and measuring the electric current intensity of electric current flowing between the first solution and the second solution over time to obtain electric current data over time; and a process of obtaining a plurality of interval time from the electric current data over time, performing a processing based on the central limit theorem regarding the plurality of interval time to obtain frequency distribution, identifying a content pattern derived from the two or more kinds of specific oligonucleotide on the basis of the frequency distribution, and determining the presence of each of the two or more kinds of specific oligonucleotide in the measurement sample from the content pattern.SELECTED DRAWING: Figure 1
【課題】2種以上の特定オリゴヌクレオチドの有無を簡便に、且つ、精度よく判定する方法を提供すること。【解決手段】2種以上の特定オリゴヌクレオチドに相補的な配列を有するプローブと測定試料とを混合し混合物を得る工程と、ナノポアを有する脂質二重膜により互いに隔てられる、前記混合物を含む第一の溶液と第二の溶液との間に電圧を印加し、前記第一の溶液と前記第二の溶液との間を流れる電流の電流強度を経時的に測定し電流経時変化データを得る工程と、前記電流経時変化データからインターバル時間を複数個求め、前記複数個のインターバル時間について中心極限定理に基づく処理を行い度数分布を得て、前記度数分布を基に前記2種以上の特定オリゴヌクレオチドに由来する含有パターンを同定し、前記含有パターンから前記測定試料における前記2種以上の特定オリゴヌクレオチドそれぞれの有無を判定する工程と、を含む判定方法である。【選択図】図1 |
