METHOD FOR QUANTIFYING 4-HYDROXYANTIPYRIN, METHOD FOR PRODUCING BIOLOGICAL COMPONENT MEASUREMENT KIT, AND BIOLOGICAL COMPONENT MEASUREMENT KIT

To provide a method for quantifying 4-hydroxyantipyrin in 4-amino antipyrin products used in a measurement kit for biological components with stable measurement accuracy without causing reduced sensitivity in an oxidase-peroxidase-coloring agent system.SOLUTION: A quantifying method of 4-hydroxyantipyrin is for quantifying 4-hydroxyantipyrin in 4-amino antipyrin products or in a reagent containing 4-amino antipyrin for colorimetric quantification of the color of a reaction solution that is used for biological component measurement kit in which hydrogen peroxide produced by the reaction of oxidase in biological components under the coexistence of peroxidase caused by oxidation and condensation of 4-amino antipyrin and hydrogen donor is generated, quantification of 4-hydroxyantipyrin is carried out by the HPLC method, and the detection sensitivity is 0.1 μg/ml or less.SELECTED DRAWING: None 【課題】 酸化酵素−ペルオキシダーゼ−発色剤系で、感度低下を生じさせることなく、安定した測定精度の生体成分の測定キットに用いられる４−アミノアンチピリン原体等における４−ヒドロキシアンチピリンを定量する方法等を提供する。【解決手段】 生体成分に酸化酵素を作用させ生成した過酸化水素が、ペルオキシダーゼの共存下、４−アミノアンチピリンと水素供与体を酸化縮合することにより生じた反応液の呈色を比色定量する生体成分測定キットに用いられる４−アミノアンチピリン原体乃至は４−アミノアンチピリンを含有する試薬中の４−ヒドロキシアンチピリンを定量する方法であって、４−ヒドロキシアンチピリンの定量をＨＰＬＣ法により行い、その検出感度が０．１μｇ／ｍｌ以下であることを特徴とする４−ヒドロキシアンチピリンの定量方法。【選択図】 なし
