Zusammenfassung: | To provide bispecific antibodies having cytotoxic activity which comprise an immunoglobulin variable region of epidermal growth factor receptor (EGFR) specific antibody as a cancer cell specific antibody, and a CD3 specific antibody immunoglobulin variable region as a T cell specific antibody, in one polypeptide chain.SOLUTION: The present invention relates to a bispecific antibody comprising an antibody polypeptide comprising a structure represented by VH1-L1-VL1-L2-VH2-L3-VL2 or the like from the N-terminal side to the C-terminal side, where VH1 represents an amino acid sequence of a heavy chain variable region of anti-EGFR immunoglobulin, VL1 represents an amino acid sequence of a light chain variable region of anti-EGFR immunoglobulin, VH2 represents an amino acid sequence of a heavy chain variable region of anti-CD3 immunoglobulin, VL2 represents an amino acid sequence of a light chain variable region of anti-CD3 immunoglobulin, as well as L1, L2 and L3 each independently represent an amino acid sequence of a linker.SELECTED DRAWING: Figure 3
【課題】癌細胞特異的抗体として上皮成長因子受容体(EGFR)特異的抗体の免疫グロブリン可変領域と、T細胞特異的抗体としてCD3特異的抗体免疫グロブリン可変領域とを一本のポリペプチド鎖に含む、細胞傷害活性を有する二重特異性抗体の提供。【解決手段】N末端側からC末端側に向かってVH1-L1-VL1-L2-VH2-L3-VL2等で表される構造を含む抗体ポリペプチドを含み、VH1は抗EGFR免疫グロブリンの重鎖可変領域のアミノ酸配列を表し、VL1は抗EGFR免疫グロブリンの軽鎖可変領域のアミノ酸配列を表し、VH2は抗CD3免疫グロブリンの重鎖可変領域のアミノ酸配列を表し、VL2は抗CD3免疫グロブリンの軽鎖可変領域のアミノ酸配列を表し、L1、L2及びL3は、それぞれ独立に、リンカーのアミノ酸配列を表す、二重特異性抗体に関する。【選択図】図3 |
