Zusammenfassung: | To provide a novel method for purifying an antibody which, in a first chromatography step of a process for purifying an antibody or an Fc-containing molecule or a substance derived from an antibody such as low molecular antibodies such as Fab, scFv, can improve the purity of the antibody as the main target substance of the affinity purification, and can also improve the selective separation properties of monomers, and can reduce or omit the burden on a subsequent impurity removal step with respect to the removal of impurities such as aggregates.SOLUTION: The invention provides a method for purifying an antibody or substance derived from an antibody characterized in that the adsorption and elution of the antibody or substance derived therefrom is carried out by using joint columns of two columns containing Carrier 1 having an affinity ligand to the antibody or substance derived therefrom and Carrier 2 having a cation exchange group respectively, or one column containing both Carrier 1 and Carrier 2.SELECTED DRAWING: Figure 2
【課題】抗体またはFc含有分子もしくはFab、scFv等の低分子化抗体等の抗体由来物質の精製工程の第一クロマトグラフィー工程において、アフィニティ精製の主要目的である抗体自体の高純度化と共に、モノマーの選択的分離特性を向上させ、凝集体等の除去に関し後段の不純物除去工程への負荷を低減または省略できる新規抗体精製手法を提供する。【解決手段】抗体または抗体由来物質に対するアフィニティリガンドを有する担体1と陽イオン交換基を有する担体2を用いて連結または混合カラムとし、一体として抗体または抗体由来物質の吸着および溶出を行うことを特徴とする抗体または抗体由来物質の精製方法。【選択図】図2 |
